Integrin-linked protein kinase (UniProt: Q13418; also known as EC:2.7.11.1, 59 kDa serine/threonine-protein kinase, Beta-integrin-linked kinase, ILK-1, ILK-2, p59ILK) is encoded by the ILK (also known as ILK1, ILK2) gene (Gene ID: 3611) in human. ILK is a member of the serine/threonine protein kinase family that is localized on the cytoplasmic side of plasma membrane and is also detected on peripheral membranes. It is highly expressed in heart followed by skeletal muscle, pancreas, and kidney. It is a receptor proximal protein kinase that regulates integrin-mediated signal transduction and serves as a mediator of inside-out integrin signaling. It undergoes rapid, but transient stimulation by cell fibronectin interactions and by insulin in a PI3-K-dependent manner, likely via the binding of PtdIns(3,4,5)P3 with a PH-like domain of ILK (aa 180-212). Its protein kinase domain is localized in amino acids 193-446 and has five ankyrin repeats within its N-terminal half. ILK is reported to phosphorylate β1 and β3 integrin subunit on serine and threonine residues. Three isoforms of ILK have been described that are produced by alternative splicing.
