α7ニコチン性受容体のアロステリックモジュレーターであるN-(5-クロロ-2,4-ジメトキシフェニル)-N'-(5-メチル-3-イソオキサゾリル)-ウレア(PNU-120596)は、アセチルコリンによって引き起こされるのと同様の立体構造変化を細胞外リガンド結合ドメインに生じさせます。PNU-120596は、α7ニコチン性アセチルコリン受容体に選択的に作用する正のアロステリックモジュレーターです。PNU-120596は、α4β2、α3β4、α9α10 nAChRによって仲介される電流に検出可能な変化を生みません。チャネルの平均の開放時間は増えますが、イオン選択性には影響しません。アゴニスト結合部位には結合しませんが、天然のエフェクターと同様の立体構造変化を誘発します。
