肝素酶 II 是肝素黄杆菌(Flavobacterium heparinum,又称肝素土地杆菌1)的三大肝素降解酶之一。它属于多糖裂解酶家族PL21。

應用

来自肝素黄杆菌(Flavobacterium heparinum)的肝素酶 II 已用于:
  • 在外泌体分离过程中消化硫酸肝素
  • 在脊索细胞条件培养基 (NCCM) 中消化硫酸肝素以研究 NCCM 中糖胺聚糖的含量
  • 用作细胞表面聚糖加工过程中消化缓冲液的组分
  • 用作肝素裂解酶(HSase)混合物,去除293ͰT/ACE2细胞表面的硫酸肝素并研究组蛋白对假病毒感染性的影响

生化/生理作用

肝素酶 II 可在己糖胺和糖醛酸残基(葡萄糖醛酸和艾杜糖醛酸)之间的α (1-4)键处裂解切割硫酸乙酰肝素,并可控制肝素裂解(相对活性约为 2:1),主要产生二糖。在三种肝素酶中,肝素酶 II 具有最广泛的底物特异性肝素酶有助于破译底物的复杂结构。肝素干扰 PCR和RNA逆转录中的DNA转录。肝素酶 II 已用于去除肝素,以进行基因组 DNA 下游分析。
