硫酸乙酰肝素蛋白多糖(HSPG)是一种糖蛋白,是基底膜不可或缺的一部分。它是由通过共价键结合硫酸肝素链的核心蛋白组成。HSPG有三种:膜HSPG、细胞外基质分泌HSPG和分泌泡蛋白多糖。

應用

硫酸乙酰肝素蛋白多糖已用于:

生化/生理作用

硫酸乙酰肝素蛋白多糖(HSPG)在多个生物过程中发挥多重功能。它们有助于形成基底膜结构并为细胞迁移提供基质。在分泌泡中,如丝甘蛋白聚糖,HSPG在颗粒状内容物的包裹、蛋白酶活性的维持、分泌后生物活性的调节方面起一定作用。HSPG可以结合和保护细胞活素、生长因子和趋化因子,有助于形成对于细胞特异性分化和白细胞募集必不可少的梯度。它们可作为蛋白酶受体和蛋白酶抑制剂,调节细胞-细胞外基质附着、细胞-细胞相互作用和细胞运动,可与生长因子受体协同作用。HSPG也可作为配体清除中的胞吞受体,并参与脂蛋白代谢和形态发生素的梯度形成。也是基质溶解因子和其他基质金属蛋白酶的对接分子。硫酸乙酰肝素可促进培养细胞附着、诱导碱性FGF与缺乏硫酸肝素和可溶性FGF受体的细胞高亲和力结合,工作浓度为10-100 ng/mL。
