應用

Sigma的软骨素酶ABC被用来消化O连接的硫酸软骨素类糖胺聚糖(CSGAG)。该酶还被用来消化蛋白质多糖中的糖胺聚糖链,在哺乳动物系统中构建和表达重组的多功能蛋白聚糖基因。此外,它还被用来消化蛋白聚糖复合物中的硫酸软骨素。该酶还被用于从蛋白聚糖中去除糖链,从而消除蛋白聚糖的轴突生长抑制作用。

生化/生理作用

软骨苷酶ABC催化含有(1-4)-β-D-己糖氨基和(1-3)-β-D-葡萄糖醛酸基(或(1-3)-α-L-iduronosyl)的多糖与含有4-脱氧-β-D-葡萄糖-4-enuronosyl基团的双糖的清除降解。它作用于硫酸软骨素4、硫酸软骨素6、硫酸皮肤素。但是,它对透明质酸盐作用缓慢。其分子量约为120 kDa,含分子量分别为86 kDa和32 kDa的两个不同亚单位。硫酸软骨素pH值为8.0,透明质酸pH值为6.8。最佳温度为37℃。1mM Zn2+ 可作为其抑制剂,0.05 M乙酸为该酶的激活剂。

準備報告

在0.01%牛血清白蛋白水溶液(BSA)中复溶。随后的稀释液可以制成pH 8.0的含有50mm Tris,60mm乙酸钠和0.02% BSA的缓冲液。溶液需新鲜配制。
