アプリケーション
脾臓1個分または細胞1~2億個分の細胞ペレットに対して1 mLの使用が推奨されます。1分間穏和に撹拌します。15~20 mLの培地または食塩水で希釈します。250~500 × gで7分間遠心し、上清をデカンテ-ションして除きます。細胞を希釈し、計数や融合用に調製します。溶解が不完全な場合は、1~4のステップを繰り返してください。
生物化学的／生理学的作用
赤血球溶解バッファ-は、ハイブリド-マ作製プロトコ-ル用に開発された製品です。融合前のマウス脾細胞浮遊液から赤血球を除去します。また、全血のような細胞浮遊液からの赤血球の除去が必要な場合にも使用できます。
構成
0.01 M Tris-HClバッファ-中に8.3 g/Lの塩化アンモニウムを含有します。

注:本製品は、マウス由来赤血球の除去を目的としています。本製品は、他の動物種の赤血球の溶解には適さない可能性があります。マウス脾細胞以外の用途に適するかどうかは、各自で決定する必要があります。
