應用
脂多糖（LPS）是革兰氏阴性细菌细胞壁的特征成分。LPS及其脂质A部分通过Toll样受体4（TLR4）刺激先天免疫系统的细胞，TLR4是Toll样受体蛋白家族的成员，该家族可识别常见的病原体相关分子模式（PAMP）。
生化／生理作用
脂多糖（LPS）位于膜的外层，并且在非包封的菌株中暴露在细胞表面上。它们有助于外膜的完整性，并保护细胞免受胆汁盐和亲脂性抗生素的作用。
準備報告
用于制备溶液的 LPS 经过苯酚提取和凝胶过滤层析纯化。通过苯酚提取制备的脂多糖含有高达 60% 的 RNA 和少于 1% 的蛋白质。随后通过凝胶过滤色谱法进行纯化，可去除苯酚提取的 LPS 中存在的大部分蛋白质，但留下的产物仍含有 10-20% 的核酸。采用离子交换色谱法进一步纯化，得到含 <1% 蛋白质和 <1% RNA 的 LPS 产品。
