Descrição geral

O dicloridrato de o-fenilenodiamina (OPD) é um substrato cromogênico que pode ser usado em procedimentos de ensaio de imunoabsorção enzimática (ELISA), pois utiliza conjugados de peroxidase de rábano. O substrato gera um produto final solúvel de cor laranja-marrom e pode ser lido por espectrofotometria em 450 nm. A reação de OPD pode ser interrompida com HCl 3N ou H2SO4 3 M e lida a 492 nm.

Reconstituição

Dissolve one tablet in 0.05 M phosphate-citrate buffer, pH 5.0, to the desired concentration (typically an OPD concentration of 0.4 mg/ml is used). Add 40 μl of fresh 30% hydrogen peroxide per 100 ml of substrate buffer solution, immediately prior to use. Tablets are individually packaged in foil packets.
