General description

Myosin-1 (UniProt: P12882; also known as Myosin heavy chain 1, Myosin heavy chain 2x, MyHC-2x, Myosin heavy chain IIx/d, MyHC-IIx/d, Myosin heavy chain skeletal muscle adult 1) is encoded by the MYH1 gene (Gene ID: 4619) in human. Myosins are highly conserved, ubiquitously expressed proteins that form regular bipolar thick filaments that together with actin thin filaments constitute the fundamental contractile unit of skeletal and cardiac muscle. Along with actin they serve as motor molecules with ATPase activity that is essential for muscle contraction. Muscle myosin is a hexameric protein that consists of 2 heavy chain subunits (MHC), 2 alkali light chain subunits (MLC) and 2 regulatory light chain subunits (MLC-2). Limited proteolysis of myosin heavy chain produces 1 light meromyosin (LMM) and 1 heavy meromyosin (HMM). HMM can be further cleaved into 2 globular sub-fragments (S1) and 1 rod-shaped sub-fragment (S2). The globular head of myosin heavy chain contains the actin-activated ATPase. It has been reported that during rest, the myosin head does interact with the actin filament due to the presence of tropomyosin. However, when muscle contracts, tropomyosin shifts out of the way to allow the globular head to engage the actin filaments. Myosin heavy chain is a conventional myosin with a N-terminal SH3-like domain (aa 37-82), a myosin motor domain (aa 86-782), and an actin-binding domain (aa 761-775). It has a rod like tail sequence that is highly repetitive, showing cycles of a 28-residue repeat pattern composed of 4 heptapeptides, characteristic for alpha-helical coiled coils
