應用

来自黑曲霉的过氧氢酶已用作:

生化/生理作用

过氧化氢酶催化过氧化氢分解为水和氧气。四聚体的每个亚基都含有与铁结合的正铁血红素IX基团。该酶还可强烈结合NADP,NADP紧邻血红素基团。测得等电点为 6.5。催化活性的最适 pH 为 7.0。酶活性受3-氨基-1-H-1,2,4-三唑、氰化物、叠氮化物、羟胺、溴化氰、2-巯基乙醇、二硫苏糖醇、联茴香胺和硝酸盐的抑制。将过氧化氢酶与抗坏血酸盐或抗坏血酸盐/Cu2+进行孵育,会导致过氧化氢酶分子的降解。
过氧化氢酶参与催化过氧化氢(H2O2)降解为水和游离氢的过程。过氧化氢酶在商业上用于分解H2O2,因为其可在工艺术流程中作为抗菌剂使用。此外,在H2O2较低浓度下,它还能催化乙醇和酚类等电子供体的氧化。过氧化氢酶通过保护细胞免受较高水平的活性氧(ROS)显示其抗氧化活性。过氧化氢酶在慢性淋巴细胞白血病、胃癌、胶质瘤和黑色系瘤中的表达会增加。过氧化氢酶在急性髓性白血病、肺癌、胰腺癌、前列腺癌和皮肤癌(非黑色素瘤)中的水平会降低。因此,过氧化氢酶在癌症中表现出双重作用。

注意

不建议冷冻溶液。

單位定義

一单位酶可在pH7.0、25℃下每分钟分解1.0 μmol的H2O2,通过测量A240的下降速率可知H2O2浓度从10.3降至9.2 mM。
