一般說明

α-酮戊二酸脱氢酶(α-KGDH,EC 1.2.4.2)是一种高度调节的Krebs循环酶,可形成线粒体2-氧戊二酸脱氢酶复合物的E1亚基。它可催化α-酮戊二酸转化为琥珀酰-CoA,并产生直接为呼吸链提供电子的NADH。降低的α-KGDH复合物活性与氧化应激和神经退行性疾病如阿尔茨海默氏症和帕金森病有关。

應用

α-酮戊二酸脱氢酶活性比色测定试剂盒已用于测定样品中α-酮戊二酸脱氢酶的酶活性。

原則

α-酮戊二酸脱氢酶活性比色测定试剂盒提供了对各种组织、细胞和分离的线粒体中α-酮戊二酸脱氢酶活性进行简单测定的方法。通过测量比色产物对α-KGDH活性进行测定,该比色产物在450nm处的吸光度(A450)与存在的酶活性成比例。在pH7.5,37℃条件下,一个单位α-酮戊二酸脱氢酶为每分钟产生1.0 μmole NADH所需的酶量。
