1,2-双十六酰基-sn-甘油-3-磷酰胆碱(DPPC)由不饱和磷脂酰胆碱(PC)通过直接的从头合成经由α1,2-二棕榈酰甘油二酯中间体而合成。磷酸胆碱转移酶酶催化1,2-二棕榈酰甘油二酯向DPPC转化。DPPC也能在重塑途径中通过不饱和PC的脱乙酰化而合成。
