Voltage-dependent calcium channel subunit alpha-2/delta-1 (UniProt: P54289; also known as Voltage-gated calcium channel subunit alpha-2/delta-1, CACNA2D1) is encoded by the CACNA2D1 (also known as CACNL2A, CCHL2A, MHS3) gene (Gene ID: 781) in human. CACNA2D1 is a dimeric protein formed by disulfide-linked 2 and chains. It regulates calcium current density and activation/inactivation kinetics of the calcium channel and is shown to play an important role in excitation-contraction coupling. It is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 25-1073), a transmembrane domain (aa 1074-1094), and a short cytoplasmic tail (aa 1095-1103). It also has a MIDAS-like motif (aa 259-263), which is present in its VWFA domain (aa 253-430). The MIDAS-like motif is shown to bind divalent metal cations and is required to promote trafficking of the a-1 (CACNA1) subunit to the plasma membrane by an integrin-like switch. Five different isoforms of CACNA2D1 have been described that are produced by alternative splicing. Isoform 1 is expressed in skeletal muscle, isoform 2 is expressed in the central nervous system, and isoforms 3, 4, and 5 are expressed in the aorta. Isoforms 2, 4, and 5 are also observed in neuroblastoma cells. Mice bearing point mutations in the CACNA2D1 gene display reduced response to pregabalin and an abnormal central nervous system synaptic transmission. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
