Quality Control Testing
Evaluated by Western Blotting in Human tonsil tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected HLA-DPB1 in Human tonsil tissue lysate.
Tested Applications
Flow Cytometry Analysis: 1 µg from a representative lot detected HLA-DPB1 in one million Human peripheral blood mononuclear cells (PBMC).
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected HLA-DPB1 in HEK 293 cells.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected HLA-DPB1 in human tonsil tissue sections.
Affinity Binding Assay: A representative lot of this antibody bound HLA-DPB1 peptide with a KD of 1.0 x 10-7 in an affinity binding assay.

HLA class II histocompatibility antigen, DP beta 1 chain (UniProt: P04440; also known as HLA class II histocompatibility antigen, DP(W4) beta chain, MHC class II antigen DPB1) is encoded by the HLA-DPB1 (also known as HLA-DP1B) gene (Gene ID: 3115) in human. HLA-DPB1 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-29), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 30-225), a transmembrane domain (aa 226-246), and a cytoplasmic domain (aa 227-258). It plays a central role in the immune system by presenting peptides derived from extracellular proteins. It is a MHC class II molecule that forms a heterodimer of α and β subunits. Its expression is observed in antigen presenting cells. It is reported to bind peptides derived from antigens that ACCESS™ the endocytic route of antigen presenting cells (APC™) and present them on the cell surface for recognition by the CD4 T-cells. Its peptide binding cleft accommodates peptides of 10-30 residues. Endocytosed exogenous antigens are readily available for presentation and compete with endogenous proteins on their way to degradation in lysosomes. Genetic variants in HLA-DPB1 are associated with increased risk of severe aplastic anemia. A mutation in HLA-DPB1 gene (HLA-DPB1Glu69) is observed in chronic beryllium disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity™, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
