Affinity Binding Assay: A representative lot of this antibody bound Raptor peptide with a KD of 5.7 x 10-6 in an affinity binding assay.

Regulatory-associated protein of mTOR (UniProt: Q8N122; also known as Raptor, p150 target of rapamycin (TOR)-scaffold protein) is encoded by the RPTOR (also known as KIAA1303, RAPTOR) gene (Gene ID: 57521) in human. Cellular mTOR is composed of two large protein complexes, known as mTORC1 and mTORC2. The mTORC1 complex is sensitive to rapamycin and consists of mTOR, Raptor, and mLST8 and two negative regulators, PRAS40 and DEPTOR. The Raptor unit serves as a scaffolding protein to bridge the interaction between mTOR and its substrates and contains seven WD repeats in its C-terminal half. AMPK is reported to inhibit mTORC1 by directly phosphorylating Raptor on serine 722 and 792. Three isoforms of Raptor have been described that are produced by alternative splicing. Isoform 1 is highly expressed in skeletal muscle, and in a lesser extent in brain, lung, small intestine, kidney, and placenta. Isoform 2 is a truncated for that lacks amino acids 380-1335. Isoform 3 is widely expressed, with highest levels in nasal mucosa and pituitary and lowest in spleen. Raptor is essential for mTORC1 activity and is critical for the regulation of mTORC1 activity in response to insulin signaling and nutrient and energy sufficiency. In addition to its role in energy metabolism and autophagy, Raptor/mTORC1 pathway is reported to play a role in lymphocyte activation through promoting 4EBP1 and S6 phosphorylation. Inhibition of Raptor/mTORC1 signaling blocks the lymphocyte expansion during primary adaptive immune response and impairs the lymphocyte mediated cytotoxic response. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Avoid repeated freeze-thaws.
