ウェデロラクトンはIκBαのリン酸化と分解をブロックすることにより、NF-κBを介した細胞の遺伝子転写を阻害します。IKKαやIKKβのキナーゼ活性を不可逆的に阻害します(IC50 < 10 μM)。p38 MAPキナーゼやAktに対する作用はありません。
