本製品は、血清型 O55:B5 の大腸菌から抽出され、イオン交換により精製されています。起源株は CDC 1644-70 です。LPS O55:B5 は、1 ng/mL でヒト腹腔マクロファージを、1～100 ng/mLでウマ腹腔マクロファージをそれぞれ刺激させるために使用されます。
アプリケーション
リポ多糖（LPS）類はグラム陰性細菌の細胞壁に特徴的な化合物です。LPSおよびそのリピド A 部分は、共通の病原体関連分子構造を認識する Toll 様受容体タンパク質ファミリー（PAMPs）の一員である Toll 様受容体 4（TLR4）により、自然免疫系細胞を刺激します。
生物化学的／生理学的作用
リポ多糖（LPS）類は膜の外層に存在し、無莢膜株では細胞表面に露出しています。リポ多糖類は外膜の完全性の維持に関与し、胆汁酸塩および脂溶性抗菌薬の作用から細胞を保護します。
調製ノート
本製品は、水（5 mg/mL）または細胞培養培地（1 mg/mL）に可溶性であり、やや濁った薄黄色の溶液を生成します。生理食塩水中でボルテックスした後70～80 oC に加熱すると、濁ってはいますがより高濃度（20 mg/mL）の溶液を調製できます。リポ多糖類は、あらゆる溶媒中でミセルを形成する分子です。水およびリン酸緩衝生理食塩水中ではやや濁った溶液を生成します。有機溶媒中でより澄明な溶液が得られるということはありません。メタノール中では浮遊物を有する混濁液が得られ、水中では均一性のやや濁った溶液が得られます。
