應用

多聚赖氨酸聚合物可用于促进细胞对固体底物的黏附、与甲氨蝶呤结合增加药物转运、胰岛的微囊化、细胞微囊化技术、微阵列载玻片包被和染色体制备等。它也被用作高效标记 和促进胞吞作用的转染剂。低分子量的多聚-L-赖氨酸 (30,000-70,000) 在溶液中粘度较低,但高分子量的版本提供了更多的每个分子的附着位点。

生化/生理作用

多聚赖氨酸是一种非特异性的粘附因子,通过增强细胞膜和培养表面带负电荷离子之间的静电作用,促进细胞与固体基质的粘附。当它被吸收到细胞培养表面时,多聚左旋赖氨酸的功能是增加细胞结合可用的带正电荷位点的编号。对于能消化多聚-L-赖氨酸的细胞,应以多聚-D-赖氨酸作为附着因子。

成分

聚-L-赖氨酸是一种带正电荷的氨基酸聚合物,每个赖氨酸残基约有一个HBr。 氢溴酸盐可使聚-L-赖氨酸呈可溶于水的结晶形式。 在β结构中可能会发现少量的产物,因为HBr干扰氨基与羧基或含N或O部分之间的氢键。

注意

无菌溶液可在2-8℃条件下稳定保存2年。脱水保存在-20°C条件下。

準備報告

聚-L-赖氨酸氢溴酸盐的分子量> 300,000。为除去 HBr,将本品溶于中性缓冲液中,透析除去盐类。未将聚-L-赖氨酸产品暴露于三氟乙酸,并通过薄层色谱法确认透析除去任何单体、二聚体或三聚体。一般以本品为附着因子,在 5 mg 聚赖氨酸中加入 50 mL 无菌组织培养级水,每 25 cm2 溶液无菌涂布表面 1 mL。5 min 后,通过抽吸清除溶液,并彻底冲洗表面。在引入细胞和培养基前干燥两小时。
