應用

多聚-L-赖氨酸聚合物可用于促进细胞与固相基质的粘附、与甲氨蝶呤结合以增加药物转运、胰岛的微囊化、细胞微囊化技术、微阵列载玻片包被和染色体制剂。低分子质量聚-L-赖氨酸(30000-70000)在溶液中粘度低,但是高分子质量版本每个分子提供更多的粘附位点。

聚-L-赖氨酸氢溴化物可用于下列应用:
  • 用于REF-52细胞实验包裹
  • 电生理实验,细胞种在涂有聚-L-赖氨酸氢溴化物或没涂的盖玻片上。
  • 大鼠神经元细胞培养
  • 单克隆抗体通过聚-L赖氨酸非直接结合

生化/生理作用

多聚-L-赖氨酸是细胞的非特异性粘附因子,它通过增强细胞膜表面的负电荷离子与培养表面之间的静电相互作用,促进细胞与固相基质的粘附。 当它被吸附到培养表面后,多聚-L-赖氨酸可增加用于细胞结合的正电荷结合位点数量。 对于能够消化多聚-L-赖氨酸的细胞,应使用多聚-D-赖氨酸作为粘附因子。

成分

聚-L-赖氨酸是一种带正电荷的氨基酸聚合物,每个赖氨酸残基约有一个HBr。 氢溴酸盐可使聚-L-赖氨酸呈可溶于水的结晶形式。 在β结构中可能会发现少量的产物,因为HBr干扰氨基与羧基或含N或O部分之间的氢键。

注意

无菌溶液可在2-8℃条件下稳定保存2年。脱水保存在-20°C条件下。

準備報告

本品的分子量为70,000-150,000。为了除去HBr,将本品溶解在中性缓冲液中并透析除去盐。 经薄层色谱法验证,多聚-L-赖氨酸未曾接触过三氟乙酸,并且已透析去除任何单体、二聚体或三聚体。 通常,本品用作粘附因子,将50 mL无菌组织培养级水加至5 mg多聚赖氨酸中,无菌条件下,每25 cm2 表面包被1 mL。 包被5分钟,然后吸去溶液并充分清表面。 晾干2小时,然后加入细胞和培养基。
