Descrição geral

O anticorpo (também conhecido como anti-α-Sm-1) é específico para a isoforma única da α-actina do músculo liso. Ele reage especificamente com a α-actina do músculo liso em ensaios de immunoblotting e marca as células de músculo liso em cortes de tecido congeladas ou embebidas em parafina e fixadas em formalina.
Actin is a highly conserved protein that is a major component of both the cytoskeletal and contractile structures in all cell types. It varies in amount, being related to the type of differentiation and to the functional state of cells and tissues. Actin can be found in two different forms of aggregation, the globular or the fibrillar state, and at least six distinct isoforms occur in vertebrates. The actins exhibit over 90% sequence homology, but each isoform has a unique NH2-terminal sequence. The isoforms are comprised of three α actins (skeletal, cardiac, smooth), one β actin (β-non-muscle) and two γ actins (γ smooth muscle and γ non-muscle).

Aplicação

A imunocitoquímica foi realizada em células do músculo liso de aortas bovinas usando o anticorpo monoclonal anti-ACTA2. As células foram cultivadas primeiro em lamínulas de vidro e fixadas em 50% de acetona/EtOH por 10 minutos a 4 graus.
Os cortes embebidos em parafina de enxertos de tecido de testículo de rato foram corados por imuno-histoquímica com anticorpo monoclonal de camundongo antimúsculo liso dirigido contra actina.
A análise por IHC do tecido cardíaco de camundongo corado com x-gal foi realizada usando o anticorpo primário, anticorpo monoclonal de camundongo antimúsculo liso dirigido contra actina, para identificar miofibroblastos.

Immunocytochemistry was performed on kertocytes fixed in 1% PFA and incubated with mouse monoclonal anti-smooth muscle actin (14A) at a 1:100 dilution.
