General description

Hermansson described the use of natural proteins immobilized onto a silica support for chiral separations in 1983 (1). Proteins contain a large number of chiral centers of one configuration, and many other sites that contribute to the general retention process. Used primarily for the separation enantiomers of basic compounds, CHIRALPAK CBH phase has cellobiohydrolase as the chiral selector immobilized on spherical 5 μm silica particles. Used in reversed-phase mode, retention and enantioselectivity is regulated by changes of pH, buffer concentration and organic modifier. The mobile phases are mixtures of phosphate or acetate buffers and organic solvents such as 2-propanol or acetonitrile. The column is preferably used for the separation of enantiomers of basic drugs, particularly compounds containing one or more nitrogen atoms along with one or more hydrogen-bonding groups (alcohol, phenol, carbonyl, amide, ether, ester, etc.).
