General description

Specificity

Clone 1C17 is a ZooMAb® rabbit recombinant monoclonal antibody that detects CHD4. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human CHD4.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell nuclear extract.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CHD4 in Raji cell nuclear extract.

Tested Applications

Western Blotting Analysis: A 1:1,000-10,000 dilution from a representative lot detected CHD4 in lysates from C6 and NIH/3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CHD4 in human stomach cancer tissue sections and human small intestine tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CHD4 in NIH/3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound with CHD4 peptide at least ten thousand-fold (10,000X) higher than with the non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Chromodomain-helicase-DNA-binding protein 4 (UniProt: Q14839; EC:3.6.4.12; also known as CHD-4, ATP-dependent helicase CHD4, Mi-2 autoantigen 218 kDa protein, Mi2-beta) is encoded by the CHD4 gene (Gene ID: 1108) in human. CHD4 is a member of the CHD family of proteins, which are characterized by the presence of chromo domains, helicase domains, and DNA-binding domains. CHD4 is a central component of the nucleosome remodeling and deacetylase (NuRD) complex, regulating gene expression, involved in early B cell development, in neuronal differentiation and the repair of DNA double-strand breaks by facilitating chromatin relaxation at damage sites. CHD4 is expressed in various tissues, with particularly high during embryonic development. CHD4 has been implicated in several diseases. Mutations and dysregulation of CHD4 are associated with various cancers, including breast, ovarian, and colorectal cancers. It is linked to intellectual disabilities and developmental delay, highlighting its importance in brain development. Aberrant CHD4 activity can lead to immune dysregulation and contribute to autoimmune diseases. CHD4 dysfunction has been associated with congenital heart defects and other cardiovascular anomalies. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
