Specificity

Clone 3F11 is a ZooMAb® Mouse recombinant monoclonal antibody that specifically detects TNF receptor-associated factor 2 (TRAF2). It targets an epitope within 15 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal half of human TNF receptor-associated factor 2 (TRAF2).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TRAF2 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TRAF2 in Jurkat cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TRAF2 in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound TRAF2 peptide with a KD of 4.2 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TRAF2 in HeLa cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

TNF receptor-associated factor 2 (UniProt: Q12933; also known as EC:2.3.2.27, E3 ubiquitin-protein ligase TRAF2, RING-type E3 ubiquitin transferase TRAF2, Tumor necrosis factor type 2 receptor-associated protein 3) is encoded by the TRAF2 (also known as TRAP3) gene (Gene ID: 7186) in human. TRAF2 is homodimeric protein that regulated the activation of  NF-kB and JNK and plays a key role in the regulation of cell survival and apoptosis. It can also form heterotrimer with TRAF1 or TRAF3. It is required for normal antibody isotype switching from IgM to IgG. It exhibits E3 ubiquitin-protein ligase activity and is reported to promote lysine 63-linked ubiquitination of target proteins, such as BIRC3, RIPK1 and TICAM1. It regulates BIRC2 and BIRC3 protein levels by inhibiting their autoubiquitination and subsequent degradation. It is an essential constituent of several E3 ubiquitin-protein ligase complexes, where it promotes the ubiquitination of target proteins by bringing them into contact with other E3 ubiquitin ligases. However, in the absence of sphingosine-1-phosphate it displays very low E3 ubiquitin ligase activity. TRAF2 has a RING-type zinc finger domain (aa 34-73) that is essential for its E3 ubiquitin ligase activity. Its coiled-coil domain (aa 299-348) mediate it homo- and hetero-oligomerization. It has two TRAF-type zinc finger domains (aa 124-180 and 177-233).  It also contains a MATH domain (aa 351-496) that binds to receptor cytoplasmic domains. TRAF2 undergoes phosphorylation at several serine residues within the first 128 amino acids. Its phosphorylation at threonine 117 is required for lysine 63- linked polyubiquitination and is also essential for its interaction with IKKα and IKKβ and subsequent activation of NF-κB. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
