Specificity

Clone 1F12-L1 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Flotillin 1. It targets an epitope within 21 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human Flotillin 1.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Flotillin 1 in HEK293 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Flotillin 1 in lysates from Rat heart tissue, and C2C12, and HCT116 cells..

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Flotillin 1 in human adrenal gland tissue sections.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected Flotillin 1 in one million Hela and HEK293 cells.

Affinity Binding Assay: A representative lot of this antibody bound Flotillin 1 with a KD of 6.2 x 10-7 in an affinity binding assay.

Target description

Flotillin-1 (UniProt: O75955; also known as FLOT-1) is encoded by the FLOT1 gene (Gene ID: 10211) in human. Flotillin-1 and -2 are evolutionary conserved, ubiquitously expressed peripheral membrane proteins that form homo- and hetero-oligomers with caveolin-1 and 2. They associate in the plasma membrane to form microdomains that have roles in cell signal transduction, regulation of cell-cell contacts, membrane-cytoskeletal interactions, and endocytosis. They may act as a scaffolding protein within caveolar membranes, functionally participating in formation of caveolae or caveolae-like vesicles. Flotillin-1 is associated with the lipid rafts and its stability is dependent on Flotillin-2. Higher levels of Flotillin-1 are reported in cases of Alzheimer s disease and its immunoreactivity has been shown in tangle-bearing neurons. However, it does not colocalize with tau protein. Flotillin-1-positive granular bodies are also found in neurons containing Pin1-positive vesicles. It is shown to accumulate in lysosomes of tangle-bearing neurons. Flotillin accumulation has also been reported in endosomal system of neurons from transgenic murine model of amyloid plaque formation and deletion of FLOT1 gene reduces the accumulation of amyloid plaques in APPPS1 mice. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
