Specificity

Clone 3F7 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects SOX-17. It targets an epitope within the C-terminal region.

Immunogen

His-tagged recombinant fragment corresponding to 149 amino acids from the C-terminal region of human SOX-17.

Application

Anti-SOX17, clone 3F7 ZooMAb, Cat. No. ZRB1345, is a recombinant Rabbit monoclonal antibody that specifically targets SOX17 and has been tested for use in Affinity Binding Assay, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected SOX17 in human fallopian tube and human cervical tissue sections.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected SOX17 in human fallopian tube and human cervical tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound recombinant SOX17 with a KD of 5.0 x 10-8 in an affinity binding assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Transcription factor SOX-17 (UniProt: Q9H6I2; also known as SOX-17) is encoded by the SOX17 gene (Gene ID: 64321) in human. SOX17 is a member of the SOX (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. It binds to the sequences 5′-AACAAT-3 or 5′-AACAAAG-3′ and modulates transcriptional regulation via WNT3A. It is reported to act upstream of the Notch system and downstream of the canonical Wnt system and provides a link between the canonical Wnt and Notch signaling. It is involved in the regulation of the formation of cells that go on to develop into endodermal tissues, such as pancreas and liver. Although detected in multiple tissues, it is preferentially expressed in esophagus, stomach and small intestine. SOX17 required for normal looping of the embryonic heart tube and in embryonic and postnatal vascular development. Inactivation of SOX17 in endothelial cells in mouse embryo is reported to cause a lack of arterial differentiation and vascular remodeling resulting in death in utero. Embryonic stem cells that are deficient in SOX17 fail to differentiate into extraembryonic cell types and maintain expression of pluripotency-associated transcription factors. However, forced expression of SOX17 results in down-regulation of embryonic stem cell associated gene expression and activates genes that are involved in differentiation toward an extraembryonic endoderm cell fate. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
