Specificity
Clone 8F3.1 is a ZooMAb® mouse recombinant monoclonal antibody that specifically detects Cyclin-dependent kinase-like 5 (CDKL5). It targets an epitope within the internal region.

Application
Quality Control Testing
Evaluated by Western Blotting in Mouse brain tissue lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected CDKL5/STK9 in Mouse brain tissue lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected c-Myc/DDK-tagged, full-length, Human recombinant CDKL5.

Target description
Cyclin-dependent kinase-like 5 (EC 2.7.11.22; UniProt O76039; also known as Serine/threonine-protein kinase 9) is encoded by the CDKL5 (also known as EIEE2, STK9) gene (Gene ID: 6792) in human. CDKL5, a proline-directed serine/threonine kinase that plays a role in regulating axon outgrowth, dendritic morphogenesis, and synapse formation in early postnatal life, and in maintaining synaptic function in the adult brain. It is detected in the nucleus and dendrites of neurons. Its expression appears to be low in the embryonic cortex and is strongly induced at perinatal and postnatal stages in maturing neurons. It is reported to shuttle between the cytoplasm and nucleus through a CRM1-mediated nuclear export mechanism. Its protein kinase domain is localized in amino acids 13-297 and the serine-threonine protein kinase active site resides in amino acids 127-144. It has large C-terminal extension of about 700 amino acids that has several functions. This region serves as a negative regulator of the catalytic activity and is also essential for its nuclear localization. Mutations in CDKL5 gene have been linked to developmental and epileptic encephalopathy 2 (DEE2), a severe form of epilepsy characterized by seizures or spasms beginning in infancy. Two isoforms of CDKL5 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
