Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Biochem/physiol Actions

UBX domain-containing protein 1 (UBXN1) recognizes breast cancer 1 (BRCA1) which is autoubiquitinated. This takes place by the binding of the ubiquitin-associated (UBA) domain of UBXN1 to the K6-linked polyubiquitin chains linked to BRCA1. The C-terminal sequences of UBXN1 also bind to the BRCA1/ BRCA1 associated RING domain 1 (BARD1) heterodimer. It then reduces the E3 ligase activity of BRCA1/BARD1 which is dependent on the ubiquitination status of BRCA1. It also functions as a negative regulator of tumor necrosis factor α (TNFα)-mediated nuclear factor kappa B (NF-κB) activation. UBXN1 interacts with the cellular inhibitors of apoptosis proteins (cIAPs) and with E3 ubiquitin ligases, which are a part of receptor-interacting protein 1 (RIP1) in the TNFα receptor complex. It tightly binds to cIAP1, blocks its recruitment to TNFR1 and inhibits the RIP1 polyubiquitination in response to TNFα.
