General description

Aldehyde dehydrogenase 18 family member A1 (ALDH18A1) is localized to the mitochondrial inner membrane. It is an ATP- and NADPH-dependent bifunctional enzyme. The gene encoding this protein is present on human chromosome 10.

Application

Biochem/physiol Actions

Aldehyde dehydrogenase 18 family member A1 (ALDH18A1) is responsible for the reduction of glutamate to δ1-pyrroline-5-carboxylate which is an important step in the de-novo biosynthesis of proline, arginine and ornithine. It is a bifunctional enzyme having both γ-glutamyl kinase (γ-GK) and γ-glutamyl phosphate reductase (γ-GPR) activities. The enzyme undergoes alternative splicing to give rise to two isoforms. The short isoform is active in the gut and it takes part in arginine biosynthesis. The long isoform is involved in the synthesis of proline from glutamate and is expressed in many tissues.
