General description

Hemagglutinin (UniProt: A0A096XG14/ A0A0R9BPQ0; also known as HA) is encoded by the HA gene (Gene ID: 3654615) in avian influenza A virus of H5N8 strain. H5N8 strain is a highly pathogenic avian influenza virus. Hemagglutinin (HA), a surface glycoprotein of influenza viruses, is a major target for detection of the influenza virus subtype in the infected samples. HA protrudes from the outside of the virus particles in the form of a trimer. It is a single-pass type I membrane protein with a single transmembrane domain that is localized in amino acids 532-554. HA is cleaved into two chains known as HA1 and HA2 by the action of TMPRSS2 or a furin-like protease of host cells. The cleavage site is localized between amino acids 345 and 346. HA binds to the sialic acid-containing receptors on the cell surface that brings about the attachment of virus to the host cell. This attachment induces virion internalization either through clathrin-dependent endocytosis or through clathrin- and caveolin-independent pathway. It can bind to animal red blood cells and cause a coagulation reaction (hemagglutination). Clone 7H6C binds to HA protein of H5N1 and H5N8, but not to HA proteins of H1N1, H3N2, and H7N9, indicating its specificity for H5-subtypes. (Ref.: Yu-Chen Cheng, Y-C., and Chang, S-C. (2020). Appl. Specificity

Clone 7H6C is a mouse monoclonal antibody that detects hemagglutinin of Avian Influenza virus, H5N8 strain. It targets an epitope within 24 amino acids from the N-terminal half.

Immunogen

His-tagged recombinant HA1 or HA2 proteins of Avian Influenza virus, H5N8 strain, expressed in E.coli.

Application

Quality Control Applications

Evaluated by Western Blotting with full-length, recombinant Hemagglutinin HA from the H5N8 strain of Avian influenza A virus.

Western Blotting Analysis (WB): A 1:500 dilution of this antibody detected full-length, recombinant Hemagglutinin HA from the H5N8 strain of Avian influenza A virus, but not the recombinant Hemagglutinin HA2.

Tested applications

Inhibition: A representative lot inhibited Hemagglutinin activity in Influenza A virus H5N8 in (Cheng, Y-C. and Chang, SC. (2021). Appl Microbiol Biotechnol. 105(1); 235-245).

Electron Microscopy: 45 g/mL from a representative lot detected Hemagglutinin, Influenza A virus H5N8 in H5/H6/H7 trivalent VLPs. (Data provided by Dr. Hui-Wen Chen (Department of Veterinary Medicine, National Taiwan University).

Immunofluorescence Analysis: 4.5 µg/mL from a representative lot detected Hemagglutinin, Influenza A virus H5N8 in Recombinant baculovirus-infected Sf21 cells (Data provided by Dr. Hui-Wen Chen (Department of Veterinary Medicine, National Taiwan University).

Western Blotting Analysis: A representative lot detected Hemagglutinin, Influenza A virus H5N8 in Western Blotting applications (Cheng, Y-C., and Chang, SC. (2021). Appl Microbiol Biotechnol. 105(1); 235-245).
