General description
An idiotype is the genetically determined variation of structures in the variable regions of immunoglobulins. Anti-idiotypic antibodies are known to target specific epitopes, on another antibody that are unique for that target antibody. The collection of epitopes for a given antibody defines its idiotype and is generally found in the complementarity determining regions of that targeted antibody. These antibodies can also bind shared epitopes on highly related monoclonal antibodies. Their specificity allows for the specific detection of antibody of interest even in the presence of endogenous polyclonal antibody. Due to their specificity for the variable region, these antibodies can also be useful in pharmacokinetic and immunogenicity studies. 564Igi is shown to be a reliable murine model of systemic lupus erythematous (SLE). These mice develop SLE profile closely resembling that of human SLE. These mice possess rearranged immunoglobulin heavy- and light-chain genes inserted into their respective loci that are together specific for nucleolar antigens. The 564 antibody is reported to have a characteristic idiotype and B cells carrying the corresponding B-cell antigen receptor are idiotype positive. The epitope recognized by the 564 idiotype antibody is a domain that may be common to multiple self-antigens. (Ref.: Berland, R., et al. (2006). Immunity 25(3); 429-440).
