Gene Information

human ... CXCL1(2919)

General description

Growth-regulated alpha protein (UniProt: P09341; also known as C-X-C motif chemokine 1, CXCL1, GRO-alpha(1-73), Melanoma growth stimulatory activity, MGSA, Neutrophil-activating protein 3, NAP-3) is encoded by the CXCL1 (also known as GRO, GRO1, GROA, MGSA, SCYB1) gene (Gene ID: 2919) in human. CXCL1 is a multi-functional chemokine that displays chemotactic activity for neutrophils. It is synthesized with a signal peptide (aa 1-34), which is subsequently cleaved off to produce the mature protein that can be further cleaved into three chains (aa 4-73; 5-73; and 6-73) via proteolytic cleavage after secretion from peripheral blood monocytes. CXCL1 may play a role in inflammation and exert its effects on endothelial cells in an autocrine fashion. Its levels are reported to be low under basal conditions but increase significantly following active infection. It plays a role in recruiting and activating neutrophils for microbial killing at the tissue site and can also activate the release of proteases and reactive oxygen species. CXCL1 signals through binding to CXCR2, a G-protein coupled receptor. Aberrant expression of CXCL1 has been reported in several cancers and has been associated with oncogenesis, metastasis, angiogenesis and chemoresistance. Clone HL2401 is shown to specifically binds to CXCL1, although it may show slight binding to CXCL2. It is shown to reduce both cellular proliferation and invasion of cancer cells overexpressing CXCL1. It also induces apoptosis and impairs tumor vasculature. (Ref.: Miyake, M., et al. (2019). Specificity

Clone HL2401 is a mouse monoclonal antibody that detects CXCL1.

Immunogen

Native human CXCL1 protein.

Application

Quality Control Testing

Evaluated by Western Blotting with recombinant Human CXCL1 protein.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected recombinant Human CXCL1 protein.

Tested Applications

ELISA Analysis: A representative lot detected CXCL1 in ELISA application (Miyake, M., et al. (2019). Theranostics. 9(3):853-867).

Inhibition Assay: A representative lot inhibited proliferation and invasion of bladder and prostate cells and disrupted endothelial sprouting in vitro. (Miyake, M., et al. (2019). Theranostics. 9(3):853-867).

Immunoprecipitation Analysis: A representative lot immunoprecipitated CXCL1 in Immunoprecipitation application (Miyake, M., et al. (2019). Theranostics. 9(3):853-867).

Western Blotting Analysis: A representative lot detected CXCL1 in Western Blotting application (Miyake, M., et al. (2019). Theranostics. 9(3):853-867).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
