General description

Nuclear factor erythroid 2-related factor 1 (NFE2-related factor 1/Nrf1), also known as NF-E2-related factor 1, NFE2-related factor 1, Nuclear factor erythroid derived 2 like 1, and encoded by the gene name Nfe2l1 and Nrf1, is a member of the bZIP family and the CNC subfamily. Isoform 3 of Nrf1 interacts with the extended kappa 3 site of the TNF alpha promoter after Fc gamma RIII stimulation and participates in the induction of this cytokine. Studies have suggested that isoform 1 of Nrf1 is either inactive or represses the transcriptional activation. Isoform 2 of Nrf1 has been shown to activate the expression of antioxidant response element (ARE) containing reporters and genes in cells, and may play a role in oxidative stress response. Other studies have suggested that isoform 1 also activates ARE-driven genes. Levels of Nrf1 isoform 2 in white fat, lung and liver are increased compared to isoform 1 (at protein level). Isoform 1 and isoform 2 of Nrf1 are widely expressed including kidney, brown fat, white fat, large intestine, small intestine, stomach, lung, brain and liver. Isoform 2 levels are elevated in brown fat and brain, but are reduced in liver compared to isoform 1 levels. Isoform 1 and isoform 2 are also expressed in mouse embryonic fibroblasts (MEF).

This Anti-NFE2-related factor 1/Nrf1 antibody is validated for use in Western Blotting for the detection of NFE2-related factor 1/Nrf1.
Western Blotting Analysis: A representative lot detected NFE2-related factor 1/Nrf1 in MEF cells stimulated with MG132 (Tsuchiya, Y., et al. (2011) Mole. Cell Biol. 31(22):4500-4512).

Quality

Evaluated by Western Blotting in MG132 treated NIH-3T3 cell lysate.

Western Blotting Analysis: 2.0 µg/mL of this antibody detected NFE2-related factor 1/Nrf1 in 10 µg of MG132 treated NIH-3T3 cell lysate.

~85 kDa observed. Uncharacterized band(s) may appear in some lysates.
