Target description

E3 ubiquitin-protein ligase RNF4 (UniProt: P78317; also known as RING finger protein 4, EC:2.3.2.27, Small nuclear ring finger protein, Protein SNURF) is encoded by the RNF4 (also known as SNURF) gene (Gene ID: 6047) in human. RNF4 gene encodes ubiquitin E3 ligase, an enzyme widely expressed in many tissues. RNF4 is mapped to human chromosome 4p16.3. The protein contains a RING-type zinc finger domain and around four small ubiquitin-related modifier (SUMO)-interaction motifs. The RING-type zinc finger domain is required for the ubiquitination of polysumoylated substrates and the SUMO interaction motifs (SIMs) mediates the binding to polysumoylated substrate. RNF4 is predominantly localized in the nucleus but can also be found in the cytoplasm. This dual localization allows it to function in multiple cellular compartments, dealing with SUMOylated substrates wherever they are located. The movement between these compartments can be regulated by specific signals or stress conditions. RNF4 is broadly expressed across various tissues, with notable levels in tissues that have high cellular turnover or are particularly responsive to stress, such as in immune cells and testis. It is involved in DNA repair, in transcription and chromatin regulation. It is also associated with the process of hematopoiesis. RNF4 is implicated in acute promyelocytic leukemia, acute myeloid leukemia, Fanconi anemia and aplastic anemia. Its role in maintaining protein homeostasis and responding to cellular stress makes it a key player in health and disease, with its dysfunction being linked to cancer, neurodegeneration, and developmental issues. Understanding the mechanisms regulating RNF4 activity and localization can provide insights into therapeutic strategies for these diseases. (Ref.: Saito, M., et al. (2014).
