General description

PAXIP1-associated glutamate-rich protein 1 (UniProt: Q9BTK6; also known as Glutamate-rich co-activator interacting with SRC1, GAS, PAXIP1-associated protein 1, PTIP-associated protein 1) is encoded by the PAGR1 (also known as C16orf53, PA1) gene (Gene ID: 79447) in human. It associates with the histone methyltransferase MLL3/4 complex, which suggests that it plays a role in epigenetic and transcriptional activation. However, in association with PAXIP1/PTIP is proposed to function at least in part independently of the MLL3/4 complex. It is proposed to be recruited by PAXIP1 to sites of DNA damage where the PAGR1:PAXIP1 complex is required for cell survival in response to DNA damage independently of the MLL3/4 complex. During immunoglobulin class switching in activated B-cells, it is involved in transcription regulation of downstream switch regions at the immunoglobulin heavy-chain (Igh) locus independently of the MLL3/4 complex. PAGR1 protein is reported to act as a transcriptional cofactor for estrogen receptor. It interacts with GR (NR3C1) and inhibits the GR transactivation.

Specificity

This polyclonal antibody detects PAXIP1-associated glutamate-rich protein 1 in human and mouse.

Immunogen

His-tagged full-length human recombinant PAXIP1-associated protein 1.

Application

Anti-PAGR1 (PA1), Cat. No. ABE1863, is a highly specific rabbit polyclonal antibody that targets PAXIP1-associated glutamate-rich protein 1 and has been tested in Chromatin Immunoprecipitation (ChIP), Immunocytochemistry, and Western Blotting.
Western Blotting Analysis: A representative lot detected PAGR1 (PA1) in Western Blotting applications (Lee, J.E., et. al. (2013). Elife. 2:e01503; Gong, Z., et. al. (2009). J Biol Chem. 284(11):7284-93).

Chromatin Immunoprecipitation Analysis: A representative lot detected PAGR1 (PA1) in MEF′s (Cho, Y.W., et. al. (2009). Cell Metab. 10(1):27-39).

Immunocytochemistry Analysis: A representative lot detected PAGR1 (PA1) in U2OS & HeLa cells (Cho, Y.W., et. al. (2009). Cell Metab. 10(1):27-39).

Quality

Evaluated by Western Blotting in HEK293 cell nuclear extract.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected PAGR1 (PA1) in 10 µg of HEK293 cell nuclear extract.
