General description
The staining method according to Leishman is one of the standard techniques in hematological diagnostic procedures. Like Wright′s eosin methylene blue solution and May-Grünwald′s eosin methylene blue solution,this ready-to-use solution is used for staining of blood and bone marrow smears and clinical cytological specimens. The typical resulting color of cell nuclei is red to purple, which depends on the buffer tablets (Buffer tablets pH 7.2 or Buffer tablets pH 6.8). The staining of the nuclei is due of the molecular interaction of the Eosin Y dye and a complex of Azur B with DNA. Both dyes assemble to an Eosin Y - Azur B-DNA complex and the intensity of the resulting stain depends on the content of Azur B and the ratio of Azur B : Eosin Y. Furthermore, the resulting stain can vary depending on the influence of fixation, staining times, pH-value of the solutions or buffer substances.
A 500 mL bottle of Leishman′s eosin methylene blue solution are sufficient for staining up to 1600 slides in Hellendahl cells. As an IVD product and CE registered, it can be used hassle-free in diagnostics and laboratory accreditations. For more details, please see instructions for use (IFU).
