Gene Information

General description

Histones are highly conserved proteins that serve as the structural scaffold for the organization of nuclear DNA into chromatin. The histones have an amino terminal tail, a globular domain, and a carboxy-terminal tail. The four core histones, H2A, H2B, H3 and H4 assemble into the octamer (2 molecules of each). Subsequently, 146 base pairs of DNA are wrapped in a sequence-independent manner around the octamer, forming the basic subunit of chromatin, the nucleosome. Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail, H4 is involved with the structure of the nucleosomes of the ′beads on a string′ structure.

Epitope: N-terminus
KLH-conjugated linear peptide corresponding to the N-terminus of human Histone H4 acetylated at Lys12.

Application

Immunocytochemistry Analysis: A 1:500 dilution from a representative lot detected acetyl Histone H4 (Lys12) in HeLa cells.

Dot Blot (Specificity) Analysis: 0.1 µg/mL from a representative lot detected acetyl Histone H4 (Lys12) in an Absurance Histone H3 Antibody Specificity Array (Cat. No. 16-667) and in an Absurance Histone H2A, H2B, H4 Antibody Specificity Array (Cat. No. 16-665).

Quality

Evaluated by Western Blotting in sodium butyrate treated HeLa acid extract.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected acetyl Histone H4 (Lys12) in 10 µg of sodium butyrate treated HeLa acid extract.
