所有胶原蛋白分子均由以三螺旋构象排列的三条肽链组成，其一级结构主要以三个氨基酸作为重复基序，即每隔两个氨基酸是一个甘氨酸残基，其前面的两个位置通常为脯氨酸或4-羟基脯氨酸。
