Specificity

Clone 2K9 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human MHC class I polypeptide-related sequence B (MICB). It targets an epitope within 20 amino acids from the N-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 20 amino acids from the N-terminal, extracellular domain of human MHC class I polypeptide-related sequence B (MICB).

Application

Anti-MICB, clone 2K9 ZooMAb, Cat No. ZRB1439, is a recombinant Rabbit monoclonal antibody that specifically targets MICB and is tested in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MICB in HeLa cell lysate.Flow Cytometry Analysis: 0.1 μg from a representative lot detected MICB in HeLa cells.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected MICB in HeLa, A431, HepG2, HUVEC, and NIH 3T3 cell lines.

Affinity Binding Assay: A representative lot of this antibody bound MICB peptide with a KD of 1.0 X 10-6 in an affinity binding assay.

Target description

MHC class I polypeptide-related sequence B (UniProt: Q29980; also known as MIC-B, MICB) is encoded by the MICB (also known as PERB11.2) gene (Gene ID: 4277) in human. MICB is a single-pass type I membrane protein that is widely expressed and acts as a stress-induced self-antigen. It is widely expressed with the exception of the central nervous system. It is also expressed in many, but not all, epithelial tumors of lung, breast, kidney, ovary, prostate and colon. It is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 23-309), a transmembrane domain (aa 310-330), and a cytoplasmic domain (aa 331-383). MICB can be induced by heat shock, oxidative stress, retinoic acid, interferon alpha, and 5-aza-2′-deoxycytidine. Its induction by interferon alpha is impaired in subjects with chronic hepatitis C virus infection. It is recognized by gamma-delta T cells and serves as ligands for the KLRK1/NKG2D receptor. Its binding to KLRK1 leads to cell lysis. Mutations in MICB gene have been linked to Rheumatoid arthritis that is characterized by inflammatory changes in the synovial membranes and articular structures, along with widespread fibrinoid degeneration of the collagen fibers in mesenchymal tissues. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in lyophilized from PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

0.3 mg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
