General description

Anti-β-Catenin-Cy3 antibody, Mouse monoclonal (mouse IgG1 isotype) is derived from the 15B8 hybridoma, produced by fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with recombinant chicken catenin. β-catenin is 92 kDa. β-catenin protein is composed of a series of protein-protein interaction motifs that allow it to function as a scaffold. The N-terminus domain contains the binding site for β-catenin as well as phosphorylation sites recognized by Glycogen synthase kinase 3 beta (GSK3β) and the C-terminus contains the transcriptional activation domain and binding site for Teashirt.
The gene β-catenin (CTNNB1) is mapped to human chromosome 3p21. The protein localizes to the cytoplasm and the nucleus. β-catenin is also present at the centrosomes.

Specificity

Recognizes specifically mammalian and avian β-catenin. Does not cross-react with plakoglobin (γ-catenin).

Suitable for direct immunofluorescence at a dilution of 1:100 using methanol/acetone-fixed bovine MDBK/MCF-7 cells. The antiobody has been used for western blot analysis in mouse embryonic stem cells.

Biochem/physiol Actions

Catenin (cadherin-associated protein) β 1 also called β-catenin is encoded by the CTNNB1 gene in humans. It is a dual function protein that regulates the coordination of cell-cell adhesion and gene transcription. It is useful for differential diagnosis of osteoblastoma (OB) and osteosarcoma (OS). β-catenin signaling pathway may contribute to cholangiocarcinoma (CCA) cell proliferation. It may serve as a prognostic marker for CCA progression and provide a potential target for CCA therapy. Mutations in β-catenin are associated with ovarian carcinoma. In addition, mutations in β-catenin are linked to intellectual disability. NEK2 (Never in mitosis A-related kinase 2) binds and phosphorylates β-catenin, thereby regulating centrosome seperation. Focal adhesion protein, Kindlin-2, enhances WNT signaling by forming a transcriptional complex with β-catenin.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
