General description

Pertussis toxin (PT), secreted protein exotoxin and virulence factor by Bordetella pertussis, is the causative agent of whooping cough. It is transported across the bacterial outer membrane through a type IV secretion system encoded by the ptl genes, situated downstream from the ptx genes that encode the toxin. This complex multisubunit toxin with an AB5 configuration, serves as a crucial component of acellular pertussis vaccines. It can function protectively as a single-component toxoid vaccine. Moreover, it is widely used in mammalian cell signaling studies due to its inhibitory effect on G-protein-coupled receptor pathways. It inhibits neutrophil recruitment and airway influx, a common inflammatory response to bacterial infection.

Application

Pertussis Toxin, Bordetella pertussis has been used as a Gi protein inhibitor to study its ability to reduce the depolarizing responses of hypothalamic pro-opiomelanocortin (POMC) neurons. It has also been used in pertussis toxin experiments to block Gαi for an in vivo inner ear and an in vitro outer hair cell (HC) study.

Physical form

Lyophilized from 50 mM NaCl, 10 mM sodium phosphate buffer, pH 7.0.

Reconstitution

Following reconstitution, refrigerate (4°C). For long-term storage in solution, reconstitute with a high ionic strength buffer, such as 100 mM sodium phosphate, 500 mM sodium chloride, pH 7.0. Stock solutions are stable for at least 6 months for use in CHO cell assays and for 1-2 months for use in non-cellular systems.
Reconstitute with 500 µl H₂O. Obtain a uniform suspension before each use by scraping the walls of the tube and gently pipetting the solution to mix.
