Quality Control Testing
Evaluated by Western Blotting in HepG2 cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected TRF4-2/PAPD5 in HepG2 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TRF4-2/PAPD5 in U2OS, NIH3T3, and L6 cell lysates.
Affinity Binding Assay: A representative lot of this antibody bound TRF4-2/PAPD5 peptide with a KD of 4.4 x 10-8 in an affinity binding assay.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TRF4-2/PAPD5 in HepG2 cells.

Terminal nucleotidyltransferase 4B (UniProt: Q8NDF8; also known as Non-canonical poly(A) RNA polymerase PAPD5, EC:2.7.7.19 PAP-associated domain-containing protein 5, Terminal guanylyltransferase, Terminal uridylyltransferase 3, TUTase 3, Topoisomerase-related function protein 4-2, TRF4-2) is encoded by the TENT4B (also known as GLD4, PAPD5, TRF4-2, TUT3) gene (Gene ID: 64282) in human. TRF4-2/PAPD5 is a component of a nucleolar TRAMP-like complex, an ATP-dependent exosome regulatory complex that consists of a helicase (MTREX), an oligadenylate polymerase (TENT4B or TENT4A), and a substrate specific RNA-binding factor (ZCCHC7 or ZCCHC8). It catalyzes preferentially the transfer of ATP and GTP on RNA 3′ poly(A) tail creating a heterogeneous 3′ poly(A) tail leading to mRNAs stabilization by protecting mRNAs from active deadenylation. It also functions as a catalytic subunit of a TRAMP-like complex that has a poly(A) RNA polymerase activity and is involved in a post-transcriptional quality control mechanism. It is shown to be essential for cytoplasmic polyadenylation of mRNAs involved in carbohydrate metabolism, including the glucose transporter SLC2A1/GLUT1. It is reported to play a role in replication-dependent histone mRNA degradation, probably through terminal uridylation of mature histone mRNAs. Five isoforms of TRF4-2/PSPD5 have been reported that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
