Clone 6G7 is a ZooMAb^® rabbit recombinant monoclonal antibody that specifically detects RNA demethylase ALKBH5. It targets an epitope within 17 amino acids from the C-terminal region. Predicted to react with all three isoforms.

Quality Control Testing

Evaluated by Western Blotting with GST-tagged human recombinant ALKBH5.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected GST-tagged human recombinant ALKBH5.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ALKBH5 in HeLa cells.

Affinity Binding Assay: A representative lot of this antibody bound ALKBH5 peptides with a KD of 2.4 x 10-7 in an affinity binding assay.

ELISA: 0.5 μg/mL from a representative lot detected human recombinant ALKBH5.

RNA demethylase ALKBH5 (UniProt: Q6P6C2; also known as EC:1.14.11.53, Alkylated DNA repair protein alkB homolog 5, Alpha-ketoglutarate-dependent dioxygenase alkB homolog 5) is encoded by the ALKBH5 (also known as ABH5, OFOXD1) gene (Gene ID: 54890) in human. ALKBH5 demethylates RNA by oxidative demethylation, specifically demethylates N6-methyladenosine (m6A) RNA, the most prevalent internal modification of mRNA in higher eukaryotes. Demethylation of m6A mRNA affects its processing and export. It is also reported to demethylate N6-methyladenosine in single-stranded DNA. For its demethylase activity, it requires molecular oxygen, alpha-ketoglutarate, and Fe2+ ions. Its alpha-ketoglutarate binding site is localized to amino acids 193-195. ALKBH5 is widely expressed, with highest expression observed in lung, followed by testis, pancreas, spleen, and ovary. Overexpression of ALKBH5 has been reported in multiple cancers, including breast cancer, glioblastoma, ovarian cancer, and gastric cancer. ALKBH5 knockout in tumor cells is shown to enhance the efficacy of immunotherapy (anti-PD-1 treatment) and prolonged survival in murine models. ALKBH5 knockout models exhibit increased m6A density and decreased mRNA expression, which leads to decreased gene expression or altered gene splicing. Some of these genes are involved in regulating VEGFa and TGFb1 in tumor microenvironment. This ZooMAb^® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
