Flow Cytometry Analysis: Staining of one million mouse splenocytes was performed using 5 μL of a 1:1 mixture of Cat. No. CWA-1032, Anti-Mouse CD5 (53-7.3) ColorWheel® Dye-Ready mAb and Cat. No. CWD-PE ColorWheel® Antibody-Ready Phycoerythrin (PE) Dye or an equivalent amount of PE-conjugated Rat IgG2a isotype control.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user

T-cell surface glycoprotein CD5 (UniProt: P13379; also known as Lymphocyte antigen 1, Ly-1, Lyt-1, CD5) is encoded by the Cd5 (also known as Ly-1) gene (Gene ID: 12507) in murine species. CD5 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 24-371), a transmembrane domain (aa 372-401), and a cytoplasmic domain (aa 402-494). In its extracellular domain it contains three scavenger receptor cysteine-rich (SRCR) domains. It is expressed on the surface of T cells bearing the β heterodimer T cell receptor (TCR) and a subset of murine B cells known as B-1a cells. It is either not present or is present at low density on the T cell subset. CD5 serves as a transmembrane receptor that regulates T cell proliferation, functions, and development. CD72 is recognized as one of the ligands for CD5. It is considered as an activation marker of T cells. Its expression is reported to increase according to the magnitude of the signal delivered by the TCR. It has been reported that the absence of CD5 in mice can enhance signaling and activation of double positive thymocytes induced by TCR stimulation, indicating that it acts as a negative regulator of TCR signaling in T cells. In mature, peripheral T cells, CD5 is reported to be present in lipid rafts of the T cell surface, where it promotes CD3 redistribution into rafts and upregulates ZAP-70 and LAT activation. Recommend storage of lyophilized product at 2-8°C. Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial. Reconstitute each vial by adding 25 μL of PBS. Protect from light.
