エステラ-ゼは、自然界に遍在していて、選択した基質に作用する一連の異なる酵素を含んでいます。91Cキットでは、クロロ酢酸ナフト-ル AS-Dが「特異的エステラ-ゼ」により酵素的加水分解され、フリ-のナフト-ル化合物を放出します。そして、ジアゾニウム化合物と複合体をつくり、酵素活性部位で濃い着色沈殿を形成します。通常、この酵素は顆粒球由来の細胞に特異的であると考えられます。末梢血、骨髄またはパラフィン包埋組織切片中の好中球を検出するのに使用されます。
