人过氧化氢酶是单功能含血红素过氧化氢酶中的一种。作为细胞内酶,它在肝脏、红细胞和肾脏中浓度较高。过氧化氢酶是同源四聚体蛋白质,由氨基酸残基、血红素基团(铁III原卟啉IX)和磷酸酰胺腺嘌呤二核苷酸(NADPH)分子组成。它是大多数需氧生物中普遍存在的酶。过氧化氢酶(CAT)基因位于人染色体11p13。

應用

来自人类红细胞的过氧化氢酶可用于:

  • 防止还原细胞色素c被H2O2再氧化,同时用细胞色素C测量超氧化物的产生
  • 作为随机光学重建显微镜(STORM)成像人类皮肤成纤维细胞成像缓冲液的组分
  • 作为单分子成像的球状混合物的组分

生化/生理作用

过氧化氢酶缺乏和氧化应激与白癜风、高血压、阿尔茨海默症、糖尿病和脱氢酶血症有关。过氧化氢酶在癌症中表现出两种作用,在慢性淋巴细胞白血病、胶质瘤、黑色素瘤和胃癌中表达水平较高,在急性髓系白血病、胰腺癌、肺癌、前列腺癌和皮肤癌(非黑色素瘤)中表达水平较低。
过氧化氢酶可激活过氧化氢(一种活性氧)分解成水和氧。它起到天然抗氧化剂的作用,可保护细胞免受蛋白质、脂质和核酸的氧化损伤。过氧化氢酶也被用于研究活性氧在基因表达和凋亡中的作用。

單位定義

在pH7.0,25℃条件下,一个单位每分钟可分解1.0 μmole的H2O2 ,H2O2的浓度从从10.3降至9.2mM(通过A240的降低速率进行测定)。
