生化/生理作用

过氧化氢酶是一种普遍存在的抗氧化酶,其功能是催化过氧化氢(H2O2) 的分解,生成水和氧气。过氧化氢在真核细胞内以多种氧化酶和超氧化歧化酶的副产物形式存在。过氧化氢在细胞内的聚集会引发细胞内DNA,蛋白质和脂质的氧化,从而导致基因突变和细胞死亡。过氧化氢酶可去除细胞内的H2O2,保护活细胞免受氧化损伤,过氧化氢酶在氧化胁迫相关疾病中的作用已得到了广泛研究。

原則

该测定方法旨在测定过氧化氢酶作用后残留的过氧化氢底物。首先,过氧化氢在过氧化氢酶作用下转化为水和氧气(过氧化氢酶途径),然后通过叠氮化钠终止这一酶促反应。随后通过比色法,分析反应混合物等分试样中余下的过氧化氢量。10 比色法使用取代苯酚(3,5-二氯-2-羟基苯磺酸),在存在过氧化氢和辣根过氧化物酶(HRP)的条件下,通过氧化作用偶联至4-氨基安替比林中,生成吸收波长520 nm的红色醌亚胺染料(N-(4-安替比林基)-3-氯-5-磺酸酯对苯醌单亚胺)。

單位定義

在pH 7.0,25℃,过氧化氢底物浓度50mM条件下,一单位过氧化氢酶每分钟可以分解 1μmol 过氧化氢生成氧气和水。
