Descrição geral
- Protocol: MS β-Glucuronidase Quick Start Guide
- MS β-Glucuronidase is a stable recombinant β-Glucuronidase mix for robust urine drug testing. Hydrolysis is completed at room-temperature or in your autosampler in a single-step . The stability and low-temperature compatibility allows an analyst to use MS β-Gluc as a bench-top reagent. Reliable hydrolysis over days on the bench means less hands-on time, less frequent reagent prep, and more automation. It has been verified against a broad spectrum of conjugated analytes, including “hard to cleave” glucuronides, particularly opiates and opioids. MS β-Gluc eliminates sample preparation bottlenecks while enabling fully automated hydrolysis and reproducible analytical results.
Aplicação
- Urine sample preparation in Forensic and Clinical toxicology laboratories
- Enzymatic hydrolysis of drug glucuronide conjugates for urine drug testing by LC/MS
- Forensic drug testing, workplace and prescription drug monitoring, antidoping, SAMHSA
- Compatible with wide range of glucuronide drug conjugates, including but not limited to opioids, opiates, benzodiazepines and cannabinoids and hormones.
Características e benefícios
- Opened bottle retains 97% activity at 25°C after 2 months.
- Stable on the bench in MasterMix (with methanol): Retains 99% activity for a week.
- Pre-buffered for use with human urine with volatile buffer ammonium acetate for LC-MS.
- Single-step hydrolysis at room-temperature or within autosampler (20-40°C).
- Verified with real human urine samples, not synthetic.
- “Walk-away” workflow.
- Automation compatibility (oven and centrifuge not required).
- Purified recombinant enzyme
Nota de preparo
For Opiates, Opioids and Broad-spectrum drug hydrolysis use 80 to 160 μL of MS ß-Glucuronidase per reaction for a 200 μL reaction in urine, so 6-12 samples can be hydrolyzed per mL of MS beta-Gluc depending on reaction temperature.
Dilute-and-shoot workflows: add MS β-glucuronidase formula and internal standards in methanol to urine and immediately transfer to autosampler at 40°C. After 16min switch temperature to 4°C. Hydrolyzed samples are ready for injection.
Workflows with extraction or filtration step: add MS β-glucuronidase formula and internal standards in methanol to urine. Incubate 30 min at room-temperature. Proceed with extraction or filtration.
