General description

YPD Broth is a nutritious general liquid growth medium commonly used for the maintenance and propagation of Pichia pastoris, Saccharomyces cerevisiae, and other yeasts. It is a medium used for molecular cloning, protein expression, or as enrichment media. Peptic digest of animal tissue provides nitrogenous nutrients. Yeast extract provides nitrogenous nutrients as well as Vitamin B Complex. Dextrose provides the carbohydrate and energy source to support the growth of S. cerevisiae.

Application

YPD Broth is recommended for the growth of Saccharomyces cerevisiae for molecular biology purposes. This medium supports the growth of most heterotrophic microorganisms but due to their simple composition, they have been adopted as the basal media for the routine cultivation of yeasts. General methods in yeast genetics specify using Yeast Extract Peptone- Dextrose (YPD) medium for cultivating S. cerevisiae and other yeasts.

Articles

The development of genetic engineering and cloning has opened many possibilities of expression and isolation of heterologous proteins for research purposes. Considerable advances in technology have enabled expression and isolation of recombinant proteins in large scale.

Technical Article on yeast media. Yeasts are eukaryotic microorganisms whose genomes have been comprehensively studied and some have been sequenced.

Protocols

Yeasts are considered model systems for eukaryotic studies as they exhibit fast growth and have dispersed cells. Yeast cultures can be grown, maintained, and stored in liquid media or on agar plates using techniques similar to those for bacterial cultures.
