Specificity

Clone 1K16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Lysozyme C. It targets an epitope within 17 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human Lysozyme C.

Application

Quality Control Testing

Evaluated by Western Blotting in HL-60 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Lysozyme C in HL-60 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Lysozyme C in Human bone marrow tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound Lysozyme C peptide with a KD of 1. 0 x 10-12 in an affinity binding assay.

Flow Cytometry Analysis: 0.1 µg from a representative lot detected Lysozyme C in one million Human peripheral blood mononuclear cells (PBMC)..

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Lysozyme C in HL-60 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Lysozyme C in Human bone marrow tissue sections.

Target description

Lysozyme C (UniProt: P61626; also known as EC:3.2.1.17, 1,4-beta-N-acetylmuramidase C) is encoded by the LYZ (also known as LZM) gene (Gene ID: 4069) in human. Lysozyme is a protein that exerts its enzymatic activity through the hydrolysis of the β-1,4-glycosidic bonds between N-acetylmuramic acid and N-acetylglucosamide in the polysaccharide backbone of the peptidoglycans of the Gram-positive bacterial cell wall. It is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature protein. Three different types of lysozymes have been described: C-type (chicken or conventional type), the G-type (goose type), and the I-type (invertebrate type). They all share the ability to hydrolyze bacterial cell wall peptidoglycan (PG) and have a similar overall structure. The C-type lysozymes are reported to be highly cationic and can kill certain bacteria independently of PG hydrolytic activity. This action may involve killing of bacteria by forming pores on their cell membrane. C-type lysozyme displays both hydrolytic and transglycosylation activities and may have a slight esterase activity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
