General description

KEAP1 is an important adapter protein important in regulating the transcription factor NFE2L2/NRF2. Nuclear factor erythroid-2 related factor 2 (NRF2) is a redox-sensitive transcription factor that positively regulates the expression of genes encoding antioxidants, xenobiotic detoxification enzymes, and drug efflux pumps, and confers cytoprotection against oxidative stress and xenobiotics in normal cells. KEAP1 negatively regulates NRF2 activity by targeting it to proteasomal degradation. KEAP1 acts as a substrate adapter protein for the E3 ligase ubiquitin protein complex formed by CUL3 and RBX1 proteins which mediate the ubiquitination and proteasome destruction of NRF2. KEAP1 also targets PGAM5 which is a central mediator of programmed necrosis induced by TNF and reactive oxygen species, so again KEAP1 plays an important role ultimately in cell survival and adaptation to its microenvironment. KEAP1 is widely expressed in many tissues, most strongly in muscle tissues and appears to shuttle from the cytoplasm to the nucleus depending upon the cellular context.

Flow Cytometry Analysis: 4.0 µg of this antibody from a representative lot detected Keap1 in 1X10E6 NIH/3T3 cells.
